No. All conversion happens locally on your Mac. Your files never leave your machine. The only exception is if you choose to upload to Google Docs (Pro feature) — and that's always your choice.
Make sure MarkDrop is running. The "Convert with MarkDrop" option appears in the Services submenu when you right-click a .md file in Finder. If it still doesn't appear, try going to System Settings > Privacy & Security > Extensions > Finder and make sure MarkDrop is enabled.
